Genetic and Environmental Influences on Blood Pressure and Serum Lipids Across Age-Groups

Aging plays a crucial role in the mechanisms of the impacts of genetic and environmental factors on blood pressure and serum lipids. However, to our knowledge, how the influence of genetic and environmental factors on the correlation between blood pressure and serum lipids changes with age remains to be determined. In this study, data from the Chinese National Twin Registry (CNTR) were used. Resting blood pressure, including systolic and diastolic blood pressure (SBP and DBP), and fasting serum lipids, including total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and triglycerides (TGs) were measured in 2378 participants (1189 twin pairs). Univariate and bivariate structural equation models examined the genetic and environmental influences on blood pressure and serum lipids among three age groups. All phenotypes showed moderate to high heritability (0.37–0.59) and moderate unique environmental variance (0.30–0.44). The heritability of all phenotypes showed a decreasing trend with age. Among all phenotypes, SBP and DBP showed a significant monotonic decreasing trend. For phenotype-phenotype pairs, the phenotypic correlation (Rph) of each pair ranged from −0.04 to 0.23, and the additive genetic correlation (Ra) ranged from 0.00 to 0.36. For TC&SBP, TC&DBP, TG&SBP and TGs&DBP, both the Rph and Ra declined with age, and the Ra difference between the young group and the older adult group is statistically significant (p < .05). The unique environmental correlation (Re) of each pair did not follow any pattern with age and remained relatively stable with age. In summary, we observed that the heritability of blood pressure was affected by age. Moreover, blood pressure and serum lipids shared common genetic backgrounds, and age had an impact on the phenotypic correlation and genetic correlations.

It is crucial to understand the genetic mechanisms and biological pathways underlying the relationship between obesity and serum lipid levels. Structural equation models (SEMs) were constructed to calculate heritability for body mass index (BMI), total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and the genetic connections between BMI and the four classes of lipids using 1197 pairs of twins from the Chinese National Twin Registry (CNTR). Bivariate genomewide association studies (GWAS) were performed to identify genetic variants associated with BMI and lipids using the records of 457 individuals, and the results were further validated in 289 individuals. The genetic background affecting BMI may differ by gender, and the heritability of males and females was 71% (95% CI [.66, .75]) and 39% (95% CI [.15, .71]) respectively. BMI was positively correlated with TC, TG and LDL-C in phenotypic and genetic correlation, while negatively correlated with HDL-C. There were gender differences in the correlation between BMI and lipids. Bivariate GWAS analysis and validation stage found 7 genes (LOC105378740, LINC02506, CSMD1, MELK, FAM81A, ERAL1 and MIR144) that were possibly related to BMI and lipid levels. The significant biological pathways were the regulation of cholesterol reverse transport and the regulation of high-density lipoprotein particle clearance (p < .001). BMI and blood lipid levels were affected by genetic factors, and they were genetically correlated. There might be gender differences in their genetic correlation. Bivariate GWAS analysis found MIR144 gene and its related biological pathways may influence obesity and lipid levels.

Background
Contrasting the well-described effects of early intervention (EI) services for youth-onset psychosis, the potential benefits of the intervention for adult-onset psychosis are uncertain. This paper aims to examine the effectiveness of EI on functioning and symptomatic improvement in adult-onset psychosis, and the optimal duration of the intervention.
Methods360 psychosis patients aged 26–55 years were randomized to receive either standard care (SC, n = 120), or case management for two (2-year EI, n = 120) or 4 years (4-year EI, n = 120) in a 4-year rater-masked, parallel-group, superiority, randomized controlled trial of treatment effectiveness (Clinicaltrials.gov: NCT00919620). Primary (i.e. social and occupational functioning) and secondary outcomes (i.e. positive and negative symptoms, and quality of life) were assessed at baseline, 6-month, and yearly for 4 years.
ResultsCompared with SC, patients with 4-year EI had better Role Functioning Scale (RFS) immediate [interaction estimate = 0.008, 95% confidence interval (CI) = 0.001–0.014, p = 0.02] and extended social network (interaction estimate = 0.011, 95% CI = 0.004–0.018, p = 0.003) scores. Specifically, these improvements were observed in the first 2 years. Compared with the 2-year EI group, the 4-year EI group had better RFS total (p = 0.01), immediate (p = 0.01), and extended social network (p = 0.05) scores at the fourth year. Meanwhile, the 4-year (p = 0.02) and 2-year EI (p = 0.004) group had less severe symptoms than the SC group at the first year.
ConclusionsSpecialized EI treatment for psychosis patients aged 26–55 should be provided for at least the initial 2 years of illness. Further treatment up to 4 years confers little benefits in this age range over the course of the study.

The aim of the present study was to compare the rate of preterm birth (PTB) and growth from birth to 18 years between twins conceived by in vitro fertilization (IVF) and twins conceived by spontaneous conception (SC) in mainland China. The retrospective cohort study included 1164 twins resulting from IVF and 25,654 twins conceived spontaneously, of which 494 from IVF and 6338 from SC were opposite-sex twins. PTB and low birth weight (LBW), and growth, including length/height and weight, were compared between the two groups at five stages: infancy (0 year), toddler period (1–2 years), preschool (3–5 years), primary or elementary school (6–11 years), and adolescence (10–18 years). Few statistically significant differences were found for LBW and growth between the two groups after adjusting for PTB and other confounders. Twins born by IVF faced an increased risk of PTB compared with those born by SC (adjusted odds ratio [aOR] 8.21, 95% confidence interval [CI] [3.19, 21.13], p < .001 in all twins and aOR 10.12, 95% CI [2.32, 44.04], p = .002 in opposite-sex twins). Twins born by IVF experienced a similar growth at five stages (0–18 years old) when compared with those born by SC. PTB risk, however, is significantly higher for twins conceived by IVF than those conceived by SC.

The objective of this study was to investigate how different obesity measures link to circulating metabolites, and whether the connections are due to genetic or environmental factors. A cross-sectional analysis was performed on follow-up survey data at the Chinese National Twin Registry (CNTR), which was conducted in four areas of China (Shandong, Jiangsu, Zhejiang and Sichuan) in 2013. The survey collected detailed questionnaire information and conducted physical examinations, fasting blood sampling and untargeted metabolomic measurements among 439 adult twins. Linear regression models and bioinformatics analysis were used to examine the relation of obesity measures, including body mass index (BMI), waist circumference (WC) and waist-to-hip ratio (WHR) with serum metabolite levels and related pathways. A co-twin control study was additionally conducted among 15 obesity-discordant monozygotic (MZ) pairs (intrapair BMI difference >3 kg/m2) to examine any differences in metabolites controlling for genetic factors. Eleven metabolites were associated with BMI, WC and WHR after controlling for genetic and shared environmental factors. Pathway analysis identified pathways such as phenylalanine metabolism, purine metabolism, valine, leucine and isoleucine biosynthesis that were associated with obesity. A wide range of unfavorable alterations in the serum metabolome was associated with obesity. Obesity-discordant twin analysis suggests that these associations are independent of genetic liability.

The Chinese National Twin Registry (CNTR), initiated in 2001, has now become the largest twin registry in Asia. From 2015 to 2018, the CNTR continued to receive Chinese government funding and had recruited 61,566 twin-pairs by 2019 to study twins discordant for specific exposures such as environmental factors, and twins discordant for disease outcomes or measures of morbidity. Omic data, including genetics, genomics, metabolomics, and proteomics, and gut microbiome will be tested. The integration of omics and digital technologies in public health will advance our understanding of precision public health. This review introduces the updates of the CNTR, including study design, sample size, biobank, zygosity assessment, advances in research and future systems epidemiologic research.
Numerical simulation of debris-removal trajectories on the transport mirrors in high-power laser systems

In high-power laser systems (HPLSs), understanding debris-removal trajectories is important in eliminating debris from the surfaces of transport mirrors online and keeping other optical components free from contamination. NS equations, the RNG $k{-}{\it\varepsilon}$ model and the discrete phase model of the Euler–Lagrange method are used to conduct numerical simulations on the trajectories of contaminant particles of different sizes and types on the mirror surface using Fluent commercial software. A useful device is fabricated based on the simulation results. This device can capture and collect debris from the mirror surface online. Consequently, the effect of debris contamination on other optical components is avoided, cleaning time is shortened, and ultimately, the cleanliness of the mirrors in HPLSs is ensured.
A novel soluble β-1,3-d-glucan Salecan reduces adiposity and improves glucose tolerance in high-fat diet-fed mice

Salecan is a recently identified water-soluble viscous extracellular β-1,3-d-glucan polysaccharide from an Agrobacterium species. It is a high-molecular-mass polymer (about 2 × 106Da) and composed of a linear chain of glucosyl residues linked through a repeat unit of seven β-(1,3) and two α-(1,3) glucosidic bonds. In the present study, we examined the effects of dietary Salecan fed at 2 and 5 % in a high-fat diet (64 % energy) in C57BL/6J mice. After 6 weeks, mice fed 2 and 5 % Salecan had significantly lower body weight, fat mass and percentage of body fat mass compared with those fed a high-fat cellulose (control) diet. Both the Salecan groups significantly and dose-dependently improved glucose tolerance, with a 9 and 26 % reduction of glucose AUC, respectively. Liver and adipose tissue weights were also significantly decreased by the Salecan treatment. Supplementation with 5 % Salecan led to lower serum TAG, total cholesterol (TC) and HDL-cholesterol (52, 18 and 19 %, respectively) and lower hepatic TAG by 56 % and TC by 22 % compared with the high-fat cellulose control group. Dietary Salecan intake caused an obvious elevation of fat in the faeces. Supplementation with Salecan disturbed bile acid-promoted emulsification and reduced the size of emulsion droplets in vitro. These results indicate that Salecan decreases fat absorption, improves glucose tolerance and has biologically important, dose-related effects on reducing high-fat diet-induced obesity.
Influence of laser surface melting on glass formation and tribological behaviors of Zr55Al10Ni5Cu30 alloy

A gradient structure was synthesized on the surface of Zr55Al10Ni5Cu30 alloy with high glass-forming ability by laser surface melting (LSM). Along the laser incident direction, the surface remelted alloy exhibits gradient microstructure distributed in the sequence of amorphous structure, nanocrystal- reinforced amorphous matrix composite (transitional layer A), dendrites–amorphous phase composite (transitional layer B), and crystalline phases from the top surface to the substrate. The formation mechanism of this gradient structure is discussed based on the experimental results of the microstructure together with the finite volume simulation of the process of LSM treatment. The friction coefficient of the transitional layer A is ∼2.5 times lower than those of the other layers under the same sliding friction condition, and possible reasons for this phenomenon are discussed in connection with the rolling motion and material transfer mechanism. The transitional layer B exhibits the best wear resistance among all the structures studied here, which is related to the optimized ratio of microhardness to reduced Young’s modulus (H/Er).
Ni- and Cu-free Zr–Al–Co–Ag bulk metallic glasses with superior glass-forming ability

Ni- and Cu-free Zr–Al–Co–Ag bulk metallic glasses (BMGs) with diameters up to 20 mm were synthesized by copper mold casting. The effects of Ag alloying on the superior glass-forming ability (GFA) of Zr–Al–Co–Ag alloys were studied based on the localized atomic structure and crystallization behavior. High-energy synchrotron radiation x-ray diffraction result reveals that Ag addition in Zr–Al–Co system results in a more homogeneous local atomic structure, which could be an origin for the improved GFA of the Zr–Al–Co–Ag alloy. Crystallization products of the Zr–Al–Co–Ag glassy alloy are more complex than those of the Zr–Al–Co glassy alloy. The Zr–Al–Co–Ag BMGs free from highly toxic elements Ni and Cu exhibited a combination of superior GFA, high compressive fracture strength over 2000 MPa, low Young’s modulus of 93 to 94 GPa, and good corrosion resistance in phosphate-buffered solution (PBS), inspiring their potential biomedical applications.

Chill-zone aluminum alloys with GPa strength and good plasticity

Using a cold graphite mold casting method, bulk AlNiY chill-zone alloys were prepared at hypereutectic compositions with Al content from 85 at.% to 94 at.%. It was found that ultra-hard surface layers with a thickness of about 200 μm and submicron grain size form when the melt can be undercooled without heterogeneous nucleation at the mold contact surface. This hard chill-zone forming in contact with the mold possesses Vickers microhardness Hv about 350–420 and is thus harder than fully amorphous Al alloys. In compression, ultimate strength more than 1.1 GPa and true strain more than 150% without failure were achieved simultaneously. The combination of high strength and good plasticity will be discussed in relation to the special structure of the chill-zone alloy.

Dendrobium Sw., a member of the family Orchidaceae, includes approximately 1100 species distributed in different parts of the world. In China, the genus is represented by 76 species and two varieties, of which D. loddigesii, D. fimbriatum, D. chrysanthum, D. officinale (=D. candidum) and D. nobile are listed in the Chinese Pharmacopoeia as the source material for Herba Dendrobii (Shihu). Because of increased demand and high price, Herba Dendrobii is often adulterated in the trade by other related species. Many Dendrobium species are over-collected from the wild and are listed under endangered taxa in the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). Therefore, a reliable authentication method is needed to regulate the trade and help conserve the species from unscrupulous collections. This review describes the present status of using medicinal Dendrobium in China, current approaches to authenticate medicinal Dendrobium plants and Herba Dendrobii, and the efforts under way towards the development of DNA microarrays to facilitate differentiation/identification of genuine material in complex Chinese medicinal formulations.

Summary
This chapter and the next examine the most commonly used methods in computational science. Here we concentrate on some basic aspects associated with numerical approximation of a function, interpolation, least-squares and spline approximations of a curve, and numerical representations of uniform and other distribution functions. We are only going to give an introductory description of these topics here as a preparation for other chapters and many of the issues will be revisited in a greater depth later. Note that some of the material covered here would require much more space if discussed thoroughly. For example, complete coverage of the issues involved in creating good random-number generators could form a separate book. Therefore, we only focus on the basics of the topics here.
Interpolation
In numerical analysis, the results obtained from computations are always approximations of the desired quantities and in most cases are within some uncertainties. This is similar to experimental observations in physics. Every single physical quantity measured carries some experimental error. We constantly encounter situations in which we need to interpolate a set of discrete data points or to fit them to an adjustable curve. It is extremely important for a physicist to be able to draw conclusions based on the information available and to generalize the knowledge gained in order to predict new phenomena.
Interpolation is needed when we want to infer some local information from a set of incomplete or discrete data.

Summary
It is usually more difficult to simulate continuous systems than discrete ones, especially when the properties under study are governed by nonlinear equations. The systems can be so complex that the length scale at the atomic level can be as important as the length scale at the macroscopic level. The basic idea in dealing with complicated systems is similar to a divide-and-conquer concept, that is, dividing the systems with an understanding of the length scales involved and then solving the problem with an appropriate method at each length scale. A specific length scale is usually associated with an energy scale, such as the average temperature of the system or the average interaction of each pair of particles. The divide-and-conquer schemes are quite powerful in a wide range of applications. However, each method has its advantages and disadvantages, depending on the particular system.
Hydrodynamic equations
In this chapter, we will discuss several methods used in simulating continuous systems. First we will discuss a quite mature method, the finite element method, which sets up the idea of partitioning the system according to physical condition. Then we will discuss another method, the particle-in-cell method, which adopts a mean-field concept in dealing with a large system involving many, many atoms, for example, 1023 atoms. This method has been very successful in the simulations of plasma, galactic, hydrodynamic, and magnetohydrodynamic systems.

Summary
The beauty of Nature is in its detail. If we are to understand different layers of scientific phenomena, tedious computations are inevitable. In the last half-century, computational approaches to many problems in science and engineering have clearly evolved into a new branch of science, computational science. With the increasing computing power of modern computers and the availability of new numerical techniques, scientists in different disciplines have started to unfold the mysteries of the so-called grand challenges, which are identified as scientific problems that will remain significant for years to come and may require teraflop computing power. These problems include, but are not limited to, global environmental modeling, virus vaccine design, and new electronic materials simulation.
Computational physics, in my view, is the foundation of computational science. It deals with basic computational problems in physics, which are closely related to the equations and computational problems in other scientific and engineering fields. For example, numerical schemes for Newton's equation can be implemented in the study of the dynamics of large molecules in chemistry and biology; algorithms for solving the Schrödinger equation are necessary in the study of electronic structures in materials science; the techniques used to solve the diffusion equation can be applied to air pollution control problems; and numerical simulations of hydrodynamic equations are needed in weather prediction and oceanic dynamics.
Important as computational physics is, it has not yet become a standard course in the curricula of many universities.

Summary
From the relevant discussions on function optimization covered in Chapters 3, 5, and 10, we by now should have realized that to find the global minimum or maximum of a multivariable function is in general a formidable task even though a search for an extreme of the same function under certain circumstances is achievable. This is the driving force behind the never-ending quest for newer and better schemes in the hope of finding a method that will ultimately lead to the discovery of the shortest path for a system to reach its overall optimal configuration.
The genetic algorithm is one of the schemes obtained from these vast efforts. The method mimics the evolution process in biology with inheritance and mutation from the parents built into the new generation as the key elements. Fitness is used as a test for maintaining a particular genetic makeup of a chromosome. The scheme was pioneered by Holland (1975) and enhanced and publicized by Goldberg (1989). Since then the scheme has been applied to many problems that involve different types of optimization processes (Bäck, Fogel, and Michalewicz, 2003). Because of its strength and potential applications in many optimization problems, we introduce the scheme and highlight some of its basic elements with a concrete example in this chapter. Several variations of the genetic algorithm have emerged in the last decade under the collective name of evolutionary algorithms and the scope of the applications has also been expanded into multi-objective optimization (Deb, 2001; Coello Coello, van Veldhuizen, and Lamont, 2002).
